When dissolving a freeze-dried peptide you decide how much water to add. That volume sets the concentration, and the concentration sets how much volume one measured amount is.
How the calculation works
The concentration is the amount of peptide divided by the volume of water. Dissolve 10 mg in 2 ml and the concentration is 5 mg per ml. Want to measure 0.25 mg from that? That is 0.05 ml, or 5 marks on a 100-unit insulin syringe.
More or less water
More water gives a lower concentration and thus a larger volume per measured amount. That makes accurate measuring easier, but the vial fills up sooner and keeps less long.
Less water gives a higher concentration and smaller volumes. That is more economical, but a small reading error on the syringe then weighs more heavily.
